As one of the most crucial Chinese medicine,the Angelica dahurica containing volatileoil, compounds such as coumarin and polysaccharides,which grows in the northeast and northChina area. The total coumarin is one of the active ingredients in Angelica dahurica,Whichwith a variety of pHarmacological effects. Angelica dahurica callus is the research object inthis study. The content of total acidity, total pHenol and total sugar in Angelica dahuricacallus were measured, the extraction process and purification process of the total coumarinin Angelica dahurica callus were be investigated, the regulation of the synthesis of the totalcoumarin in Angelica dahurica callus were studied. These afford a scientific basis forcomprehensive exploitation and application of Angelica dahurica callus.1、The content of total acidity, total pHenol and total sugar in Angelica dahurica callus weremeasured, and the content was respectively1.261%、0.334%and0.662%. These components accounted for about2.257%in Angelica dahurica callus, and thatchoosing an appropriate extraction and separated methods to remove these impurities couldprovide the foundation basis for the separation and purification of the total coumarin.2、Single factor test and the orthogonal test were used to to extract the total coumarin inAngelica dahurica callus. The optimizing the extraction process was respectively as follows:ethanol solution60%, pH9, extraction temperature70℃, extraction time30min. There was agood linear relationship within the range of3.0~30.0μg/mL (R=0.999). The averagerecovery was98.03%(n=3)with RSD of2.4%(n=10).3、D-101macro porous resin was chosen as a resin for purification of thetotal coumarin. Single factor test and the orthogonal test were used to optimize itspurification process,which was that adsorption flow rate of0.5mL/min, eluent methanolconcentration of95%, eluent volume of9BV volume,elution rate of0.5mL/min, and theaverage content of coumarin was78.73%, the purity was72.71%.4、In order to study basic composition and ion concentration of culture medium on the growth of Angelica dahurica callus and effects of the total coumarin synthesis process. The resultsshowed that the appropriate medium for the grows of Angelica dahurica callus was: MS+TDZ1.5mg/L+NAA0.2mg/L,the appropriate medium for the accumulation of the totalcoumarin was: MS+TDZ1.5mg/L+NAA0.2mg/LIn this paper, the UV spectropHotometric method for the determination of total coumarinfrom Angelica dahurica callus was established.The parameters of ultrasonic extraction andpurification and separation of macro porous resin were obtained, And discusses the factorsthat affects the changes of total coumarin content in Angelica dahurica callus, and providethe basis for the industrialization production and pHarmacological research in Angelicadahurica callus.
