Bioactive peptide is a kind of peptides compound, which has the physiological action for the life activity of biological organisms. With the deep research of structures and functions, more than one hundred kinds of peptides have been found and separated from human body by scientists, and more features have been known by people, which makes the study of the area has experienced unprecedented prosperity. Bioactive peptides have become the research hot spot of biological field, and been widely applied in the medical field, especially the study of peptide drugs, and its importance will be more and more significantly.The new bioactive peptides LS-7and PR-6used in the issue are obtaind through the bioinformatics softwares (provided by tutor), and predicted which have the activity of inhibiting cancer cell growth., and are not yet available related sequences and functional activity reported. This study is order to establish a set of new solid-phase peptide synthesis scheme of LS-7and PR-6. We discussed the optimization of conditions, that is the choice of resin and deblock time, and then the function of inhibiting cancer cell growth activity will be verified initially.The LS-7and PR-6were synthesiszed by Solid-Phase method. This method utilized Wang resin as supporter, Fmoc-amino acid as the raw material, DCC/HOBt as coupling reagents and95%TFA/H2O as the cleavage reagents. It was identified by ESI-MS that the synthesized peptide have the same mass weight with the theoretical one, which confirmed that the new bioactive peptide LS-7and PR-6were synthesized successfully. After the crude products of LS-7and PR-6were separated and purified by reversed-phase high performance liquid chromatography, the purity was risen from22.36%and70.40%to92.39%and93.32%respectively. And then the activity of the purified products were determined invitro. The leukemia cells HL-60, liver cancer cells HepG2and gastric cancer cells BGC-832were treated by these peptides in RPMI1640medium with96-cell plate in5×104/well respectively. Finally, the rate of inhibition were detected by the MTT in570nm. To research the inhibitory effect on leukemic cells of two kinds of peptides, PR-6inhibition effect on leukemic cells was significantly, and the rate could reach47.9%at the concentration of0.2μM. However, the inhibitory effect of LS-7was lower than PR-6, and the rate could only reach21.3%at the concentration of0.1μM. To research the inhibitory effect on of liver cancer cells and gastric cancer cells, the action effect of LS-7on gastric cancer cells was better than liver cancer cells at the concentration of0.1μM, while at the concentration of0.01μM the effection on liver cancer cells is better. the activity of inhibiting cancer cell growth.Through the research, the synthetic schemes of biopeptides by chemical synthesis is feasible, the effect experiments show that the peptides have good activity of inhibiting cancer cells growth, and it provides a new type of molecular and functional evidence for the development of peptide drugs.
