With the comparison of the effects under gradient concentration Cu2+ condition, the Tea poly-phenol oxidase enzyme synthesis Thea-flavins efficiency. The determination of tea samples polyphenol oxidase is utilized in modern extraction and separation technology, which has been analyzed by HPLC oxidation product. The results show that, in this test the synthesis efficiency under gradients concentration Cu2+ condition varies linearly.1. The results show the optimal value for tea leaves appears in the condition of [Cu2+/PPO]=0.2, the ethyl acetate layer total Thea-flavin content of 45.738%, with a synthesis rate of 35.193%. However, in [Cu2+/PPO] =0.8 condition, Tea poly-phenol oxidase oxidation effect back to the status of [Cu2+/PPO]=0. When [Cu2+/PPO]>0.8, Cu2+ appears on poly-phenols oxidase inhibition. On the other side, conversion rate on catechin, when [Cu2+/PPO]=0.4, the maximum conversion rate up to 86.80%, after [Cu2+/PPO]>0.4, catechin conversion rate declined. The condition of [Cu2+/PPO]=0.2 is the optimal value for tea poly-phenol oxidase enzymatic oxidation synthesizing.2. The results show the optimal value for apple fruit appears in the condition of [Cu2+/PPO]=0.2, the ethyl acetate layer total Thea-flavin content of 19.125%, with a synthesis rate of 13.345%. However, in [Cu2+/PPO] =0.8 condition, Tea poly-phenol oxidase oxidation effect back to the status of [Cu2+/PPO]=0. When [Cu2+/PPO]>0.8, Cu2+ appears on poly-phenols oxidase inhibition. On the other side, conversion rate on catechin, when [Cu2+/PPO]=0.2, the maximum conversion rate up to 99.05%, after [Cu2+/PPO]>0.2, catechin conversion rate declined. The condition of [Cu2+/PPO]=0.2 is the optimal value for tea poly-phenol oxidase enzymatic oxidation synthesizing.3. The results show the optimal value for potato earth-nut appears in the condition of [Cu2+/PPO]=0.2, the ethyl acetate layer total Thea-flavin content of 36.909%, with a synthesis rate of 22.022%. However, in [Cu2+/PPO] =0.8 condition, Tea poly-phenol oxidase oxidation effect back to the status of [Cu2+/PPO]=0. When [Cu2+/PPO]>0.8, Cu2+ appears on poly-phenols oxidase inhibition. On the other side, conversion rate on catechin, when [Cu2+/PPO]=0.4, the maximum conversion rate up to 94.72%, after [Cu2+/PPO]>0.2, catechin conversion rate declined. The condition of [Cu2+/PPO]=0.2 is the optimal value for tea poly-phenol oxidase enzymatic oxidation synthesizing.
