Recent years, the secondary metabolites of higher fungus have been attractting more and more chemical and biological researchers.A large number of chemical constituents with novel skeleton were found in higher fungus, some of which possessing significant bioactivities have been developed to drugs or lead compounds.Thus, the metabolites of Cyathus striatus have been extracted and separated, and their chemical constituents have been identified, their bioactivities have been tested. By using repeated chromatographic column on silica gel, Sephadex LH-20, revesed phase C18 and HPLC, 10 compounds, including 6 new cyathane diterpenoids were obtained from the large scale fermentation cultures of the microbial materials. The structures were elucidated on the basis of spectroscopic analysis(1H-NMR、13C-NMR、HSQC、1H-1H COSY、HMBC、NOESY and MS). The results are concluded as follows:Six cyathane diterpenoids have been seperated from the fermentation broth of Cyathus striatus: striatoids A-F(1-6). Among them, both 2 and 3 carried an unusual epoxy bridge between carbons 15 and 4’, and this is the first report in this class of natural products. Four known metabolites,including cyclic dipeptides and benzene derivatives:Cyclo-(S-Pro-R-Leu)(7), 2-Hydroxymethyl-phenol(8),(2R,3S)-pterosin C(9),(2S,3S-pterosin C(10).Among them, Compounds 1-6 were tested for their enhancement of NGF-induced neutrite outgrowth in PC12 cells. All the compounds All the six compounds dose-dependently enhanced neurite formation.
