Apoptosis is the process of cell independent death under the regulation of gene in order to maintain the body’s normal development and homeostasis, which happens throughout the whole life of an organism. Apoptosis plays an important role for the organization update, embryonic development, immune and other physiological mechanisms operation, abnormal regulation of apoptosis will cause cancer, myocardial infarction, neurodegenerative diseases and many other diseases, the study of apoptosis is of great importance in the field of life sciences and clinical medicine. Quick and accurate apoptosis detection is necessary and basal for cell apoptosis in-depth study, the clinical diagnosis of the disease also has a strong demand for the convenience of the detection, as the traditional methods have complex operations, large equipment and other reasons, the present research attempts to design a simple, portable apoptosis detection devices.Microfluidic chip is a kind of highly interdisciplinary micro analysis system which can manipulate, separate samples. Combining with electrical, optical, fluid dynamics and other technical means would make microfluidic chips achieve the aim to separate and detect cells, proteins, nucleic acids and other biological components accurately and rapidly. Dielectrophoresis technique is suitable to control cells and other neutral particles which can be polarized in microscale, cells exhibit different motion characteristics due to their different dielectric properties in the electric field. On account of cell dielectric properties, this study used the electrorotaion microfluidic chip which based on dielectrophoresis technology to detect cell apoptosis.The project designed and produced a kind of electric rotary chip, designed the chip control system and set up a test platform, researchers detected out of the cell apoptosis in different periods have different rotational spectral characteristics, differentiating the apoptotic cells successfully. The main results are as follows:1. A kind of electric rotary microfluidic chip was designed and producted based on PCB materias. The sf9 suspension cells were tested with the chip of type 300μmsix-electrodes, under 8V applied voltage, among 10~250KHz frequency range, the cellsrotated clockwise.2. We produced a kind of electric rotary chip signal controller and built the electric rotating test platform. The controller can output continuously adjustable voltage of2.5~15V, output continuously adjustable frequency 0~200V, it can switch the 4/6 pole of the chip, the controller can also adjust an electric field for the cell chip for simple positioning and capture; the electric rotating test platform includes a signal generation system, the signal control systems, the signal detection system and the image acquisition system.3. Glass material was used to improve a electrical rotating chip which specifications is 200μm-6-electrode. Under the control of conditions of 8V voltage,250 KHz frequency, conductivity of extracellular suspensions under 500μS/cm, the Hela cells of normal hydrogen peroxide-induced apoptosis show the significant difference of rotational speed, verified the feasibility to use electric rotary chip theory to detecte apoptosis.
