Using the transgenic insect-resistant maize Bt-799 and its non-transgenic parent as materials, a sandwich enzyme-linked immunosorbent assay(ELISA) method was applied to measure the content of mCrylAc protein in different growth stages and different tissues of the transgenic Bt-mCrylAc maize, to investigated the agronomic traits of maize and comparison of Bt-mCrylAc gene influence on absorption of nutrition elements of maize plants, Study on feed safety of transgenic Bt-mCry1Ac maize Bt-799 through feeding zebrafish, to provide the basic data for health and environment, safety evaluation of transgenic Bt Maize. The main research contents and results are as following:1.the Bt-mCrylAc can express in leaves、stems、roots and seeds of the maize throughout the entire growth period. The expression level of the Bt gene showed significant temporal and spatial patterns in different tissues during different growth stages of Bt-799; the content of Bt-mCrylAc protein in the maize had an evident increase from the tillering stage to seed maturation, and reached the peak at the seed maturing stage; meanwhile, the content of Bt-mCrylAc protein showed significant difference among various plant organs, and was significantly higher in leave samples than other organs, and was the lowest (2.86±1.71μg/g) in the seed samples.2.Transgenic Bt-mCrylAc maize Bt-799 and its non-transgenic near isogenic lines of Zheng 58 in the same period of plant height, ear height, tasseling, silking stage have no significant difference.3.Through the determination of transgenic Bt-mCry1Ac maize Bt-799 and its non-transgenic near isogenic lines of Zheng 58 in different period of nutrition and total nitrogen, total phosphorus, total potassium of root, stem, leaf, The results show that two kinds of corn nutrient elements exist a certain differences. Compared with non transgenic Zheng 58, Transgenic Bt-mCrylAc maize Bt-799 roots and leaves in the different stages of the total P uptake have no significant difference, in the seedling stage, jointing stage and anthesis absorption, there are no significant difference between the two kinds of corn on the absorption of total nitrogen and total potassium. However, there are significant differences in grain filling period and mature period in two corns.4. With Bt-mCrylAc transgenic maize Bt-799 and its non transgenic near isogenic lines of Zheng 58, a 14-weeks zebrafish feeding tril, the results showed that the growth indexes, organ coefficient and oviposition are no significant differences between Bt group expanded feed and non-Bt group expanded feed. All the growth parameters, and egg laying organ coefficient has no significant difference, between Bt corn powder group and non-Bt groupm aize powder.
