In Quaternary Period, the global climate change has a great effect on the growth and evolution of the existing species, their geographical distribution, as well as population genetic structure. In recent years, the emergence and development of phylogeography have provided an effective way to conjecture the shelter of the existing species in Quaternary glaciation. At present, our research on phylogeography is focused on regions which are located in southwestern, southern and northwestern of China. However, rare research has been made on the shelter of various species of plants in northeastern China during the Quaternary glaciation. To further study the shelter and expanding route of some main plants in northeastern China during the Quaternary glaciation, my research will place Adonis amurensis Regel et Radde as a research object, which can lay a theoretical foundation for the evolutionary distribution pattern of modern plants in this area and meanwhile for the protection of biodiversity. On the study, we used the chloroplast DNA universal primers rps16R2-rps16 F research the phylogeography of Adonis amurensis. We got the haplotype network, geographical distribution of haplotypes and phylogenetic tree of Adonis amurensis. Based on the study of 143 samples which were seleted from 18 population, we noted that the populations which Located in the eastern part of jilin possess high genetic diversity. DH(Hd = 0.711, π = 0.0012)、LSH(Hd = 0.571, π = 0.0019)、JY(Hd = 0.536, π =0.0019)、JTJ(Hd = 0.533, π = 0.0021). Through the AMOVA analysis of cp DNA data, we noted that the genetic variation of Adonis amurensis mainly exist in the population. In addition, combined the haplotype network with geographical distribution, 17 populations(in addition to GH) can be divided into three major regions- northeast and southeast of Hei Longjiang, eastern of Ji Lin and southeastern Liao Ning. As can be seen from the FST value, the lagest genetic distance(0.51156) exists in the populations of Heilongjiang and Liaoning. Therefore, the populations which are located in the eastern of Ji Lin have richer haplotypes, the region may be the diffusion center of Adonis amurensis in the interglacial, and also is a biological refuge in the LGM. Based on the results, we speculated that the north of Changbai Mountain is probablya the refuge of Adonis amurensis in the Quaternary glacial. This conclusion is consistent with Qian’s view.
