Cellulose is a kind of high polymer compound, the molecular formula is (C6H10O5)n,and the range from150,000to350,000weight average molecular weight, which is equivalentto300~15000glucose medium. Cellulose is closely linked with people’s life and hasimportant social value, medicinal value, economic value and scientific research value, so itwidely used in food, medicine, papermaking, chemical, building materials, plastics,explosives and electrical etc. China, with a thousand years history of farming civilizationagricultural, widely planted flax, wheat, rice and sugarcane, coupled with our vast forestresources, which greatly enrich our country cellulose material sources. At present, amountsfor textile and paper cellulose reach to8million tons per year in the whole world. Becausecellulose is insoluble in water and common organic solvents, so most cellulose resource havenot been effective use and after used is difficult to degrade, which will bring enormouspressure to environmental protection.With above questions, this study isolated a strain and optimization of its fermentationsystem to promote gradually increased degradation or circular cellulose and reduce theenvironment pollution. This paper selected the regions in Changli county of Hebei province asthe sampling. The object is extracting and separating and strain screening from soil samplesthe through a filter paper medium and carboxymethyl cellulose agar, and then this workanalyzed and discussed the factors influencing the activities of the cellulose. Hope this workcan provide the theory basis for research related to properties of cellulose, literature cellulosedegradation bacteria and make due contributions to the construction of ecologicalenvironment protection in china. The main results are as follows:1. Different soil samples from different soil sampleThe distribution of cellulose degrading bacteria in soil samples is different, there isobvious difference. Acquisition point selection has a direct effect on bacteria and itsscreening.2. Screening for target strains.Screening one cellulolytic fungi and marked as M4. It was preliminary inferred astrichoderma viride. M4can use and degradation of carboxymethyl cellulose, carboxymethylcellulose and good growth in the plate, and the filter paper strip rapid disintegration.3Optimization of enzyme-production ability of bacteriumOptimization of fermentation conditions for experiments showed that the best carbonsource, culture medium was CMC-Na,1.5%; the optimum nitrogen source for yeast extractwas0.5%; the optimum liquid volume was50mL; the optimum fermentation temperature was35℃; The optimum enzyme production time was third days; the optimum. 4. Influence factors of activities of the cellulase(I) the optimum pH is5.5for M4enzymatic reaction; the optimum reaction time was46min; enzymatic reaction temperature is wider, higher values in the range of35~60℃forenzyme activity, however in the range of55~60℃the enzyme activity was the highest, theoptimum temperature is55℃for enzymatic reaction.(II) The concentration of metal ions have an important influence on the enzymaticreaction. Fe2+, Cu2+at the concentration of20~50mg L-1has significant activation on theenzymatic reaction and Mg2+activation are generally.(III) There is no positive correlation between oscillation speed and cellulose activity.When the speed is high enzyme activity decreases and cause side effects.
