As we all know,the arctic is a unique geographicai location,which create a unique ecological environment in arctic,for example, cold,long frozen time,strong radiation and so on.The arctic has become a hot spot which scientists eage to research in all over world. For unique ecological environment in arctic,the arctic microorganisms has unique molecular biology mechanism and physiological and biochemical characteristics that adapt to the arctic environment. arctic microorganisms also has become a hot research field.This thesis mainly focus on diversity of the arctic ocean microorganisms,In the past, researcher mainly focus on bacteria diversity of the arctic sea ice and polar ocean sediments,but research on diversity of the culturable bacteria in arctic sea is rare.We simulate the bacteria living environment of arctic location,and cultivate culturable bacteria in arctic sea.Then we bulild16SrDNA library studing the diversity of culturable bacteria in the arctic ocean,analysis evolution relationship of culturable bacteria,reveal the difference between culturable bacteria groups in different regions and different depth in arctic ocean.This study enrich the16SrDNA library of arctic ocean bacteria, provide basic molecular biology data to understand diversity of culturable bacteria in the arctic ocean,and also provide some theoretical basis for the development and utilization of the polar arctic microorganisms.In this study,we bulild16SrDNA library to analysis diversity of culturable bacteriain in five station in different regions and different depth in arctic ocean (B80-40m station, B80-3.5km station, B79-100m station, C19station, B84A-30m station).The results show diversity of culturable bacteria in the Arctic marine is rich,there is some culturable bacteria which can not be identified in all five different stations.for example, more than60%clonings can not be identified in B80-40m station.All above reveal that huge microbial resources exist in arctic sea. The phylogenetic tree show that the culturable bacteria which can be identified is belong to three groups, the donminant group is Alphaproteobacteria, followed by Gammaproteobacteria and Cytophagia.In five stations. the culturable bacteria which can be identified can be be divided into15bacteria groups, Hyphomonas, Roseobacter, Parvularculaceae, Alpha proteobacterium, Metlycophaga, Gamme proteobacter, Flexibacter, Marinomonas, Pseudomonas, Brevundimonas, Sphingomonas, Providencia, Alcaligenes, Vibrio, Zobelia.We screen101strains bacteria from arctic sample, sequences analysis shows101strainsbacteria is belong to Alphaproteobacteria,Gammaproteobacteria and Cytophagia.this result is consistent with the result of diversity of culturable bacteria above.In five stations.101strains bacteria can be be divided into10bacteria groups:Glaciecola、Roseobacte、 Halomonas、Rhodococcus、Kocuria、Brevundimonas、Psychrobacter、 Pseudorhodobacter、Sphingomonas、Flexibacter.
