With the increase of throughput and speed and decrease of cost for the next generation sequencing technology, we can sequence bacterium genome quickly, and provide rich information for genome comparisons. We made genetical studies on none-pathogenic symbiosis intestinal tract Escherichia coli of human and vertebrate. The goal is to discover the genetical differences bewteen Escherichia coli in intestinal tract of healthy Human and Vertebrate, to explain the genetically foundation of the different relation between E. coli and it’s host.We sequenced a draft genome of E. coli in healthy chinese person intestinal tract(designated E. coli H001), and another draft genome of E. coli in healthy Mexico Alouatta palliata intestinal tract(designated E. coli V001), then made genome annotation and comparision, explained genes and genetical elements in significance.We used Solexa GAII genome sequencer, adopted multiplex method to make Pair end sequencing, each read contains80x2bp, placed E. coli H001and E. coli V001and other3bacteria in the same lane to sequence and use6mer Index strategy to identify sequence of the two strains. Used SOAPdenovo software to assembly the genome denovo, used Mauve software to make syntenic comparison between the sequenced strains and reference strains. We used GLIMMER software to identify ORFs, used InterProscan and BLAST to annotate the genome seperately. We made Circular representation of E. coli H001and V001strain to gain the general characters of the genome. We used WEGO software to make Gene Ontology to classify the genomes functionally, identify and explain the unique genes of the genomes.As result of the sequencing and analysis, E. coli H001has a draft genome of4,766,659bp, GC content is50.9%, harbors154Scaffolds, encode4670predictive genes,964putative proteins. E. coli V001has a draft genome of4,738,010bp, GC content is50.5%, harbors49Scaffolds, encode4646predictive genes,1095putative proteins.E. coli H001strain has more gene to promote genetic variation and have more genomic variations, both strains has beta-lactam resistance genes. E. coli H001strain has more genes to support benzene metobolism and ion homeostasis, E. coli V001strain has more genes for nutrient substance transport. The human and the vertebrate have a different living environment, food and drink structure and social structure, so the Escherichia coli in there intestinal tract have different traits.
