Life events, biological evolution and diseases are the significant studies of oursociety. To fully understand organism, firstly we should know more about basicactivity of organism, that’s gene regulation. The study of gene regulation always wasthe most important theme. Gene regulation is the basic measure for organismaccommodating to the environment. Studying more about gene regulation can helpus to understand the diversity and adaptability of the organism. To compare thedifference among gene regulations of different lives can help us to study theevolution of lives. Also, utilizing the theory of gene regulation, some diseases causedby gene mutation can be cured.There are many kinds of gene regulation in the organism. We can study themfrom the basic gene regulation. I study the Lac self-regulating genetic circuit in thispaper. I study the circuit from its protein expression. I designed a pure circuit withoutthe effect of DNA segment in conventional circuit. To eliminate effect of multi-copythe circuit was conformed into chromosome. The study of basic gene regulation canexplain complex gene regulation and organism eventsIn chapter one, I give a conceptional explain of the gene regulation. Then Iintroduce the specificity and control point of gene regulation, and the basic elementsof transcriptional activation in gene regulation. At last, I give different kinds offluorescent proteins because of experiment demand.In the second chapter, I analyze the theory and principles in experimentsincluding basic principle of self-repression, theoretical calculations about proteinexpression of self-repressing circuit in statistics, and regulation theory and basicmodel of Lac circuit’s design.The third chapter analyzes biological method demanded in experiments. Itintroduces the design process of Lac genetic circuit. Then it introduces the measuringmethod. I use Flow Cytometry to measure the distribution of protein expression. Theresult can verify the theoretical calculations.At the end, I summarize the paper and give the prospect of the paper. I putforward the progressive significance of this paper.
