Lutein is an important natural food colorant and additive, which is also effective in delaying chronic disease.Lutein is widely used in food, cosmetics, health products and other fields. Lutein has great market value and wide application prospect. Micro algae belongs to the plant, most of them are single-celled plants. Micro algae can produce lutein, which is very valuable in economic and environmental sense. The micro algae is used to produce biodiesel, lutein is an important kind of high value-added products as well. It’s very necessary that researching in the extraction conditions of lutein.In this study, two methods are used in extracting lutein and optimizing the extraction condition. Studying ultrasonic assisted extraction technology, the ultrasonic time and power, solid-liquid ratio, and extraction agent are optimized, when the ultrasonic power is300W, solid-liquid ratio is1:25, ultrasonic time is10min (ultrasonic10s, intermittent10s), We get the higher extraction; Studying digestion auxiliary extraction process, optimize the usage amount, digestion time, stirring time of cellulase, xylanase, and pectinase. When cellulase and xylanase by volume ratio is3:2, pH5.0, and reaction time is2.5h, reaction temperature is50℃, magnetic blender mixing time is1.5h, we obtain the highest content of lutein, which increased21.38%than using a single enzyme.In the condition that25%sodium hydroxide-ethanol solution as the medium of saponification crude extract, saponifier volume ratio of4:1, saponification time1.5h, saponification temperature is50℃, lutein get high content and purity.
