Enzyme are biological macromolecules with biological catalytic function. According to its chemical composition, it can be divided into two large categories: protein enzyme(P) and nucleic acid enzyme(R). Organisms contain lots of different kinds of enzymes, the coordinating role of certain enzyme constitutes the metabolic pathway of organisms, and metabolic pathway is foundation to sustain life. Different enzymes have their specific half-life, but the enzyme in the most important “metabolic control” general have a short half-life. Enzymes in organisms are constantly in the process of upgrading, to make enzymes in a dynamic equilibrium.In this paper, we study the hydrolysis enzyme, like alpha amylase(aspergillus oryzae, bacillus licheniformis) and invertase(saccharomyces cerevisiae), Amyloglucosidase(aspergillus Niger), trypsin(bovine pancreas), protease K(Tritirachium album limber) and so on. Research the trypsin interaction with alpha amylase by Zeta potential, and alpha amylase, invertase, Amyloglucosidase enzyme hydrolysis its specificity substrate, we can get the following main conclusions:(1) The surface of enzyme exist Zeta potential. under the same buffer system, different enzymes have different Zeta potential, and in the process of enzymatic reaction, Zeta potential changes;(2) Under the same conditions, when the enzyme and substrate have the same Zeta potential, reduce the Zeta potential of substrate can improve the response speed of enzymatic reaction;(3) In the process of enzymatic reaction, part of the peptide bond in the enzyme will disconnect, and the free amino acids gradually appear in the reaction system. a) The amino acid fall off the enzyme in enzymatic reaction is a common phenomenon. Take alpha amylase for example, the free amino acids have more than ten kinds, and the main amino acids are threonine, serine and glutamin acid;b) Within a certain range of substrate concentration, amino acids and substrate concentration are positively correlated; c) The amino acids fall off the enzyme after enzymatic reaction may be one of reasons for the half-life.
