Chalcone synthase is a key enzyme metabolic pathways from benzene propane metabolism to flavonoids synthesis.Its catalytic 3 malonyl Co A with 2 molecules coumatic acyl Co A generated 4,5,7-thihydroxy flavanone(narigenin chalcone). The grapes contains extremely rich in nutrients, including polyphenols. It is a mass of fruits and often used to make juice, raisins and wine. But the resistance of grape is poorer, vulnerable to environmental impact, resulting in a decline in its production and quality. Plants have immune function resistance self defense mechanism that similar to the animal.To explore CHS function in grape regulation and metabolic pathways of secondary metabolism transformation, the experiment research as follows:1.Two chalcone synthase genes were cloned by using RT- PCR method from Vitis vinifera. The open reading frames(ORF) of two CHS c DNAs were named Vv CHS1 and Vv CHS3 respectively. The sequence of Vv CHS1 is 1182 bp, coding 393 amino acids and the sequence of Vv CHS3 is 1170 bp, encoding 389 amino acids.2.In the TIGR gene index database, use graoe and chalcone synthase for keyword,find out all Vv CHS EST query, then using NCBI ORF finder prediction program, get 5 complete ORF sequences, and take the bioinformatics analysis. Including:the level of amino acid structure; physical and chemical properties; hydrophobicity; secondary structure; tertiary structure and evolution analysis.3.After the grape leaf were treated under different stresses UV and SA, we analyzed the changes of relative expression levels of Vv CHS1 and Vv CHS3, by Real-time PCR method. The results showed that, the stress of different processing conditions, the gene relative expression quantity are changed, the tendency of slow down after rising first. This suggests that Vv CHS1 and Vv CHS3 participate in plant resistance.In this paper, analysis the Vv CHS gene and its coding prorein, laid a foundation for the study the function of Vv CHS gene and its role in grape adaptive.
