Chemical investigation on S. thermophilum YM6-1cultured in YGP led to the purification and identification of three known metabolites,including6-hydroxymellein (1), orthosporin (2) and4,8-dihydroxy-3,4-dihydronaphthalen-1(2H)-one (3). This is the first report on6-hydroxymellein (1) and orthosporin (2) secondary metabolites isolated from Scytalidium sp.HPLC and LC-MS indicated that the contents of arthrosporol A in macroporous resin50%methanol fraction of ethyl acetate extracts of ΔAOL_s00083g86mutant was higher than wild-type andΔAOLs00078g383mutant. The result indicated the sesquiterpenes cyclase genes AOL_s00078g383and AOL_s00083g86were not directly involved in the biosynthesis of arthrosporols in A. oligospora. HPLC results showed that the ΔAOL_s00083g86mutant tended to produce more diverse compounds than ΔAOL_s00078g383.Chemical investigation on fermentation broths of AAOL_s00078g383and ΔAOL_s00083g86mutants led to isolation of five metabolites (4-8), including two new hybrid PKS/TPS compounds (7-8), as well as three known metabolites:tyrosol (5), arthrosporol A (6) and4-hydroxy-3-methoxyphenylethanol (4).
