Glutamine synthetase (GS) a key ensyme of ammonium assimilation. This paper used the model plant Arabidopsis thaliana of the glutamine synthetase gene as the object of study, the GS1.2 and GS1.4 expression in response to salt stress characteristics, analysis GS1.2 and GS1.4, trying to reveal the effect of salt stress tolerance in Arabidopsis thaliana.Firstly, by means of semi quantitative and Northern blot hybridization, expression of WT GS1.2 and GS1.4 gene and different abiotic environmental stress and different nitrogen source processing conditions under and in WT GS1.2 and GS1.4 gene expression quantity were studied. Research results show that, gene GS1.2 and GS1.4 has significant spatial and temporal expression specificity. The two genes seeds are not expressed in stem; GS1.2 in seedling stage expression amount is greater than the germination period expression, and GS1.4 in germination period expression than that of seedling stage expression; GS1.2 mainly in roots and flowers have abundantly expressed GS1.4 mainly in roots and leaves of mature expression. GS1.2 GS1.4 during seed germination stage and seedling stage of salt stress response relationship exists, all of which were up-regulated, suggesting that GS1.2 GS1.4 for salt stress stress related genes; two weeks of seedling, cadmium stress treatment, and GS1.4 GS1.2 showed marked up regulation of expression; KNO3 and urea as nitrogen source, GS1.4 and GS1.2 also up-regulated expression, NH4C1, GS1.2 is down regulated expression, and GS1.4 no response was significantly.Secondly, by inflorescence infection method constructs the expression plant Arabidopsis GS1.2 and GS1.4 gene of the screening breeding obtain T3 homozygous lines, and were identified by Southern and Northern blot methods, the three independent lines. Wild type and GS1.2 and GS1.4 expression in plants under salt stress treatment results show that GS1.2 and GS1.4 over expression plants growth were better than that of the wild type, and the salt stress treatment, overexpression of plant root length is slightly longer than the wild type, the overexpression of GS 1.2 and GS1.4 were the seeds under salt coercion germination rate has increased, so it is inferred that overexpression of GS1.2 and GS1.4 gene can enhance the tolerance of Arabidopsis plants to salt stress stress.To sum up, GS1.4 and GS1.2 has significant spatial and temporal expression specificity, and response to different abiotic environmental stress and nitrogen source, the overexpression of GS1.2 and GS1.4 gene can improve the resistance of Arabidopsis plants to salt stress stress.
