In this paper, experimental study conducted freeze-thaw broken, ultrasonic broken, high pressure homogenization broken, freeze-thaw and ultrasonic joint broken, freeze-thaw and high pressure homogenization joint broken. To explore the pros and cons of various methods broken with algae blue protein purity and yield as the metrics. The results show that:During the freeze-thaw broken experiment, with increase of moisture content, purity and yield slowly lower. Purity and yield declined after a period of stability with increase of freeze temperature. Increase the frequency of freeze-thaw, purity and yield significantly reduced after increased firstly. The result showed that the optional medium is PBS buffer solution, the concentration is 0.75%(W/V).During the ultrasonic broken experiment, with increase of moisture content, purity and yield lower. The solvent extraction is solution of CaCl2, the concentration is 0.375%(W/V). With increase of broken time, purity decreased slowly after the highest value added, and yield slowly increased. Purity and yield decreased constantly after improved significantly with increase of broken power. Purity and yield decreased constantly with increase of broken frequency.During the high pressure homogenization broken experiment, purity and yield increased with increase of moisture content. The solvent extraction is buffer solution of PBS, the concentration is 0.75%(W/V). Increase the homogenization pressure, purity and yield reduced after increased. Purity and yield reduced quickly after increased with increase of homogenization frequency.During the freeze-thaw and ultrasonic joint broken experiment, with deionized water as the extraction solvent extraction has no effect on phycocyanin extraction. Studies have shown that the joint methods of ultrasonic brokening after freeze-thawing one time can be the best selection. The solvent extraction is solution of CaCl2, the concentration is 0.25%(W/V), and the moisture content of blue algae is 96.5%. And ultrasonic brokening one time in 6 minutes by 1W power is the setting conditions.During the freeze-thaw and high pressure homogenization joint broken experiment, with deionized water or PBS buffer solution as extraction solvent have a promoting effect on phycocyanin extraction. Studies have shown that the joint methods of high pressure homogenization brokening after freeze-thawing one time can be the best selection. The solvent extraction is buffer solution of PBS, the concentration is 0.5%(W/V), and the moisture content of blue algae is 98%. And high pressure homogenization brokening one time by 4Mpa pressure is the setting conditions.
