The process of plant growth and development was induced by internal and external factors. Cytokinin play an important role in cell division and tissue development in plant growth and development. And the cytokinins responsive promoter can provide a convenient study method for analyzing the distribution of cytokinins in plant organs or tissues.We obtained the Arabidopsis response regulators promoter of cytokinin (ARR5) from Arabidopsis thaliana genome by using PCR technology.As the proteins encoded by the ARR5 gene response to cytokinins, the ARR5 promoter were fused to the beta-glucuronidase (GUS) gene and the plant expression vector pBI101-ARR5-GUS was built in this experiment. Then the fused gene was introduced via Agrobacterium tumefaciens into the genome of Crassula arborescens and Arabidopsis. The young foliage pieces of Crassula arborescens explants were chosn to study regeneration and genetic transformation. The transgenic Crassula arborescens and Arabidopsis plants obtained by Agrobacterium tumefaciens-mediated method We analyzed the best concentration of Agrobacterium for infection was investigated in this study. And resistant seedlings of Crassula arborescens and Arabidopsis were obtained and then confirmed by PCR identification. In addition, the result of GUS histochemical staining of showed the successful fusion of ARR5 promoter and GUS gene. This study will provide materials for the further study of the distribution of cytokinins during Crassula arborescens differentiation and Arabidopsis senescence.
