In the present study, xylanase was immobilized by chitosan, alginate and D280 macroporous resin, then we optimized the process conditions and did research on the enzymatic property of immobilized xylanase. The results are as follows:1. Immobilized xylanase was prepared successfully by chitosan, the single-factor experiment determined the best process conditions was: the amount of enzyme was 450 U, p H was 5.8, process-time was 5h, temperature was 40℃; orthogonal experiment got the best process conditions was:the amount of enzyme was 450 U, p H was 5.6, process-time was 5h,temperature was 40℃. Meanwhile, we got the highest recovery of enzyme activity, was 35.6%;2. Immobilized xylanase was prepared successfully by alginate, the single-factor experiment determined the best process conditions was: the amount of enzyme was 360 U, p H was 5.8, process-time was 3h, temperature was 30℃; orthogonal experiment got the best process conditions was:the amount of enzyme was 360 U, p H was 5.6, process-time was 3h,temperature was 30℃. Meanwhile, we got the highest recovery of enzyme activity, was 39.3%;3. Immobilized xylanase was prepared successfully by D280 macroporous resin, the single-factor experiment determined the best process conditions was: the amount of enzyme was 450 U, p H was 5.3, process-time was 7h,temperature was 50℃; orthogonal experiment determined the best process conditions was:the amount of enzyme was 450 U, p H was 5.1, process-time was 8h, temperature was 52℃, Meanwhile, we got the highest recovery of enzyme activity, was 40.8%;4. We analyzed the enzymatic property of immobilized xylanase by three kinds of material,the results showed that the three kinds of immobilized xylanase was stronger than free enzyme on storage stability, heat stability and p H stability, and the D280 was the best than the other two;5. The kinetics of enzyme-catalyzed reactions of immobilized xylanase was compared with free enzyme, the three kinds of immobilized xylanase had the hihger Km and lower Vmax, but the D280 was similar to free enzyme, it was high affinity for substrate.In conclusion,The xylanase Immobilized by D280 macroporous resin as carrier was superior to chitosan and alginate.
