There are many human therapeutic proteins using industrial bioreactors to produce, but the establishment of such a system is time-consuming and relatively expensive. As people’s growing demand for pharmaceutical proteins, manufacture a economical and efficient production of pharmaceutical proteins present bioreactor has become a hot research. With the rapid development of bio-pharmaceutical industry,transgenic chicken oviduct bioreactor is becoming the focus of attention, With the excellence advantage,transgenic chicken will benew industry andone of hot spot of Scientific research inthe field of biological pharmacy industry.The most successful method to make transgenic chicken is Retrovirus mediated method right now,but there has no system report about technology platform to make transgenic chicken with Retrovirus.Based on this experiment in the laboratory before,the principle of efficientto make appropriate improvements fortransgenic chicken production methods. First synthesized human epidermal growth factor plasmid,we used has made progresses on OV promoter, modification after transcription and so on. the steps to produce virus is the same, the concentration and titration of virus have approved and using reagent box to ensure maximum activity of the virus.the absolute quantification PCR we used can determinate the virus’s copy number accuratly at RNA level. And we got the titers that about 7× 107copies/ml. After obtained virus,infection with the virus chicken embryo fibroblasts, Do mention protein found in cells can express hEGF protein by Westernblot. Wemake needle by machinewhich ensure the syringe smooth rather than rough,this can minimize the hurt to the embryo. In the open aspect of our eggs are hot sol, eggshell film better method to improve the egg hatchability,eventually up to 43 percent hatchability. Finally, we have designed the individual aspects of gene and protein detection PCR assay to detect transfer, PCR detection of transgenic efficiency can reach 46%. Achieve goals efficiently produce transgenic chickens.
