Estrogen plays crucial roles in development of ovary and uterus, and neurotrophic factors also take part in animal reproduction. Recent studies have demonstrated that neurotrophins have effects on the secretion of estrogen. However, little is known about the effects of estrogen on expression of neurotrophins. This study took 21-day-old female mice as research object, treating them for a week with 17β-estrogen, then analyze the expression of neurotrophins in ovaries and uteruses The results as follows:1. The expression of BDNF and its receptor Trk B in porcine COCs.The study found that mice weight increment slowed down and ovaries and uteruses weight increased by long-term and high dosage treatment of estrogen. The estrogen period of mice were not regular, most of them were keep in metestrus. By Hematoxylin-Eosin(HE), we observed that small antral follicles quantity increased significantly, uterus endometrium and outer membrane thickened, uterus induced edema.2. Expression of BDNF, NGF, NT-3 and NT-4/5 by estrogen in ovaries.The results of RT-PCR indicated that the expression of BDNF and NGF in ovaries of mice treated with 17β-estrogen have a significant dcrease, the expression of NT-3 had a significant decrease, and the expression of NT-4/5 had no change. The results of Western-Blot showed an accordant trend with that of RT-PCR.3. Expression of BDNF and NGF by estrogen in uteruses.The results of RT-PCR indicated that the expression of BDNF in uterus of mice treated with 17β-estrogen have a significant increase, while the expression of NGF had a significant decrease. The results of Western-Blot showed an accordant trend with that of RT-PCR.By summarizing the above results, Long-term and high dosage treatment of estrogen produce an effect on form and expression of neurotrophins in ovaries and uteruses of mice.
