In recent years, an ageing population in developed countries is deepening year by year, even in developing countries such as China. The old people’s physical quality is bad, some age-related diseases such as cancer, alzheimer’s disease incidence is increased. In order to achieve the prevention and treatment of age-related diseases, to delay aging and prolong life, the purpose of the research on the mechanisms of aging increasingly becoming the concerned focus of today’s life science and one of the main challenge. The study on aging and anti-aging is becoming one of hot spots and growing points in the field of life sciences. Growth differentiation factor 11 (GDF11) is a newly identified anti-aging factor, which shows a definite anti-aging activity in mammalian species. However, It remains largely unclear how GDF11 to influence lifespan. In recent years, the annual freshwater egg-laying N. Guentheri get more and more get the favour of aging researchers. Compared with vertebrates shuch as mice, zebrafish, N. Guentheri because of its short life is suitable for aging research.In this study, a cDNA for GDF11 was cloned from the.N. Guentheri. The cDNA we obtained contains an open reading frame (ORF) of 1170 bp codes for a deduced protein of 390 amino acids. GDF11 gene with the signal peptide, TGF-β propeptide and the TGFGF-β domain. Sequence comparison revealed that the predicted protein was 97.2% identical with N. Guentheri GDF11 and Sparus aurata GDF11. N. Guentheri GDF11 is 71.2%,71.2%,70.4%,71.2%,79.3% and 72.9% identical to GDFllof human, mouse, wild boar, rabbit, crocodiles and tropical clawed frog, respectively. In this study, Real time Quantit PCR showed that the transcripts of 6-month N. Guentheri GDF11 and 9-month N. Guentheri GDF11 are specifically expressed in brain and liver, with weak expression in heart, gill, kidney, skin, bowel, gonad and muscles.In conclusion, the GDF.1.1 gene was cloned from the annual fish Nothobranchius guentheri. We analyze the sequence and structure of GDF11, results showing that the genomic structure, protein domain and 3D molecular models of GDF11 were analogous to GDF11 in human and other mammals. GDF11 is expressed in the various tissues of N. Guentheri, with the most abundant expression in brain. Our study on relationship between age and expression in N. Guentheri shows that GDF11 gene expression (except expression in brain and liver) decrease with aggravating aging process. This study laid the foundation for molecular mechanism for N. Guentheri GDF11 anti-aging research.
