Purpose Bacterial wilt of the eggpant was a kind of soil-borne disease that was infected by Ralstonia solanacearum, which caused the plant rapidly withered, it is a main reason of causing yield reduction in eggplant. Some previous researches show that AP2/ERF transcription factors play an important role in plant’s growth, development and environmental stress, such as cold, hot, saline-alkali and disease stresses. Our research separated and identified mixed Ralstonia solanacearum. We via bioinformatics analyze show that there were 130 AP2/ERF family members in eggplant and researched the mechanism of ERF transcription factors in Ralstonia solanacearum resistance through tissue expressions, oxidative stresses and differential expression patterns under Ralstonia solanacearum treatment on resistant and susceptible varieties. In order to provides the theory basis in via gene engineering and molecular biological method for functional genes in Ralstonia solanacearum resistance.Method In this study, applications of molecular markers of ISSR appraised 34 kinds of Ralstonia solanacearum’s kinship. Isolation and identification of eggplant of the ERF transcription factor by used of bioinformatics, verified the oxidative stress, tissue expression and anti-infected vaccination varieties R. solanacearum differential of expression by q PCR,builted Silence System of VIGS basis on PDS. Used of VIGS to analysis 8 Sm ERF gene’s phenotypic of silence and evaluate disease-resistant.Results 1.We study of 34 kinds of Ralstonia strains isolated diseased plants through ISSR molecular markers and genetic diversity show that distance ralstonia strains are genetically similar to the 34 kinds, it can be broadly divided into four categories. 39 and 40 is substantially similar to R. solanacearum strains at the genetic evolution, is considered to be the same strain. 2.Bioinformatics methods eggplant AP2 / ERF transcription factor family members, distribution, structure and function to analysis. The results showed that: eggplant AP2 / ERF family includes 130 proteins, including AP2, RAV, ERF subfamily each containing 20, 2 and 108 members, ERF divided DREB / CBF(41 members) and ERF(67 members) two large subfamily Phylogenetic analysis indicated that both divided into 11 small subfamilies. AP2 / ERF contains 20 important motifs, where the amino acids conserved domains on the same subfamily ERF member protein sequences constitute similar, suggesting that the presence of these motifs for the implementation of ERF protein function is required: Indicates Sm ERF transcription factor motif analysis.3. q PCR for 38 Sm ERF gene tissue expression analysis showed that the presence of tissue Sm ERF differentially expressed. 33 Sm ERF genes by heat, cold, salt, heavy metals cadmium, SA and ABA induced stress treatments. Which induced high and low temperature rendering complementary phenomena; consistent gene expression under salt stress treatment with cadmium condition trend; gene expression ABA and SA treatment similar. The results show that the ERF transcription factors involved in defense under the conditions of eggplant Stress responses. The results of this study will provide the theoretical basis for in-depth study eggplant AP2 / ERF family genes, lay the foundation for further analytic AP2 / ERF gene function. 4.After inbred speciest Kanggan R. solanacearum bacterial wilt differential expression analysis showed that: Sm ERF34, Sm ERF39, Sm ERF60, Sm ERF62, Sm ERF80, Sm ERF88, Sm ERF100 and other 24 genes associated with resistance to bacterial wilt might eggplant. 5.Established based on the PDS(phytoene dehydrogenase) VIGS silencing system and constructed the presence of Ralstonia solanacearum treatment conditions significantly differentially expressed genes VIGS vectors, expression analysis and silence inoculated found Sm ERF60,Sm ERF88 participation eggplant resistant wilt process.Conclusion Our study was analyzed Ralstonia solanacearum in the level of evolution in order to providing data support for research of Ralstonia solanacearum resistance in eggplant. It is the first time to launch bioinformatics analyzing in ERF transcription factors family of eggplant and illustrated the compartmentalization of the members and families of transcription factors in eggplant. The oxidative stresses, hormonal stresses and tissue expressions analyzing indicated that ERF transcription factors participated the defense responses of eggplant under different stress conditions. There were differential resistances on resistant and susceptible varieties of eggplant after ERF transcription factors silence under Ralstonia solanacearum treatment. It is in order to researched the functions of ERF transcription factors of eggplant in Ralstonia solanacearum resistance.
