MARVELD1 locates on human chromosome 10q24.2, its full-length mRNA is 3238 bp, encoding a 18.9 kDa protein, it has multiple biological functions such as inhibiting tumor proliferation, cell cycle arrest, etc. Our previous research found that the human and mouse source MARVLED1 have high degree of similarity, we analyzed the MARVELD1 expression levels by using in situ hybridization in mouse embryos. We found m MARVELD1 was mainly expressed in the brain and endoderm organs in E12.5~E15.5 mouse embryos, for example the small intestine. The data suggests MARVELD1 may also play an important role in mouse development.To further explore the role of MARVELD1 in mouse development, especially in intestine. We used Cre/loxp recombination system to produced MARVELD1 knockout mice. In addition, small intestine is an ideal biological model of self-renewal and homeostasis, thus, this research selects MARVELD1 knockout mouse’s small intestine as the main object. We compared the small intestine between postnatal MARVELD1 knockout mice and wild-type C57BL/6J mice by using immunofluorescence, immunohistochemistry staining and a variety of methods. This research mainly pay attention to postnatal MARVELD1 knockout mice’s weight, the length of the small intestine, small intestine histological change and various cell types change of small intestine.Experimental results showed that wild-type mice and knockout mice has no significant difference in basic signs such as drinking, eating, body hair characteristics. Compared with wild-type C57BL/6J mice’s body weight, postnatal MARVELD1 knockout mice have lighter weight to 31 days, and the maximum difference is in 41 day, p=0.06. Meanwhile, MARVELD1 knockout mice’s small intestine length have no significant with wild-type C57BL/6J mice’s small intestine length. Importantly, MARVELD1 knockout mice’s small intestine appear striated border and grassroots breakable defective, low protein expression of villin, a significant reduction in the number of goblet cells and Paneth cells.In conclusion, comparing with wild type mice, postnatal MARVELD1 knockout mice’s histological structure and some functional cells of the small intestine change.
