1403C, a structurally novel anthraquinone derivative, has been isolated from cultures of a mangrove endophytic fungus No.1403 and proved to be a proming anti-tumor compound, with mildest cytotoxim effects on normal cells. However, the production of 1403C was quite low. Therefore, the fermentation process of mangrove endophytic fungus was investigated and optimized from the following three aspects:optimization of the fermentation process and pulse fed-batch strageties, scale-up of the fermentation process.It was found that mangrove endophytic fungus No.1403 was a aerobic filamentous fungus. Controlling fermentation broth pH around 7.0 during the first 24 h of the fermentation process was preferable for the mycelial growth, but not for the biosynthesis of 1403C. After fermentation 48 h, began to control fermentation broth pH around 7.0, which was favourable for the biosynthesis of 1403C.Different pulse fed-batch strategies indicated that the initial glucose concentration was significant for the mycelial growth and the biosynthesis of 1403C. When the initial glucose concentration was 10 g/L, pulse fed-batch was not preferable for the biosynthesis of 1403C, with feeding malonic acid, the production of 1403C was enhanced markedly, and the yield of 1403C reached 1.96 g/L, which was 148%of the batch fermentation in 5 L bioreactor. When the initial glucose concentration was 10 g/L, pulse fed-batch was not only preferable for the mycelial growth, but also observably improved the production of 1403C, the dry cell weight reached 4.5 g/L, the production of 1403C reached 2.64 g/L, the yield coefficient of Y1403c/x reached 0.59 g/g DCW, which was 122.6%,198.5%,163.9%of the batch cultivation in 5 L bioreactor respectively. Carbon and nitrogen mixture feeding extended the period of biosynthesis 1403C, the dry cell weight reached 4.7 g/L, the production of 1403C reached 2.46 g/L, the yield coefficient of Y1403C/x reached 0.52 g/g DCW, which was 128.1%,184.9%, 144.4%of the batch cultivation in 5 L bioreactor respectively.Finally, the scale-up of 1403C fermentation process was successfully. The production of 1403C reached 1.54 g/L for batch fermentation in 30 L bioreactor. The production of 1403C reached 1.93 g/L for batch fermentation in 500 L bioreactor...
