Synthesis, Characterization And Study On Antitumor Activites Of New Type Platinum(Ⅳ) Complexes

In recent years, the anticancer activity of platinum complex has been more and moreattention by people. Its anticancer mechanism is generally considered to be the platinum drugswith DNA to form covalent or intrastrand cross-linking. But its structure of trans-platinum,multi-core platinum and monofunctional platinum is not different from cisplatin compounds,so generally not with DNAto form a similar intrastrand cross-linking, but with DNAto frominterstrand cross-linking, and long-interstrand cross-linking or single-function adduct.In order to overcome the shortcomings of cisplatin toxicity and solubility, designed andsynthesized a mixed amine platinum （Ⅳ） complexes （amine include: ammonia molecules,aniline, cyclohexylamine, pyridine, etc.）: Pt(C₆H₁₁NH₂)₂I₂,[Pt(C₆H₁₁NH₂)₂I₂]₂, Pt(C₆H₁₁NH₂)-（C₆H₅NH₂）I₂, Pt(C₆H₁₁NH₂)（CH₃C₆H₄NH₂）I₂, Pt(C₆H₁₁NH₂)（C₅H₅N）I₂, Pt(C₆H₁₁NH₂)-（CH₃C₅H₄N）I₂, Pt(C₆H₁₁NH₂)（NH₃）I₂, Pt(C₆H₁₁NH₂)（C₆H₅NH₂）（COOCH₃）₄, Pt(C₆H₁₁NH₂)-（CH₃C₆H₄NH₂）（COOCH₃）₄, Pt(C₆H₁₁NH₂)（C₅H₅N）（COOCH₃）₄, Pt(C₆H₁₁NH₂)（CH₃C₅H₄N）-（COOCH₃）₄and Pt(C₆H₁₁NH₂)（NH₃）（COOCH₃）₄. Platinum complexes of ammine are quitepromising against some cancer according to the literatures. At the same time, we alreadysynthesized seven precursor complexes of platinum and five platinum complexes for the firsttime. They have been characterized by elemental analyses1H nuclear magnetic resonance andso on.The complexes that have been characterized were anti-tumor activity screening. By MTTmethod, the complexes have been assayed for cytotoxicity in vitro against four cell lines：HL-60, BGC-823, KB, and Bel-7402. By contrast cisplatin, this paper discusses the structureof the complexes of the cytotoxicity of influence. The results indicated that complex3,5nothave good cytotoxicity against Bel-7402、HL-60、BGC-823and KB. Complexes3relativelyobtained better cytotoxicity against Bel-7402，but compared with cisplatin is not goodcytotoxicity.The final chapter in our laboratory conditions, using the spectrophotometric method toexplore the determination of nitrite in water. H₂O₂-Rhodamine B catalytic spectrophotometricdetermination of nitrite in water samples under the optimized conditions, nitrite has a good linear relationship （r>0.999）, the linear range of0.1³.0ug/mL. The wide linear range ofthis method can be used with pre-treatment determination of Nitrite more substance.
