Comparative Study On Different Extraction Method Of Tussah Pupa Skin Chitosan

Chitin in terms of food, health, sanitation, health and so on, has a very practicaleffect. Chitosan is a deacetylated product of chitin, it also known as soluble chitin.Because chitosan higher than chitin biocompatibility, it is more suitable to be used bythe body to absorb. Tussah pupa skin is reeling industry and food industry wastes, itchitin-rich, so that about30%. Full utilization and development of chitin and chitosan,have very important economic and social value.Traditional tussah pupa skin chitosan extraction process, is still limited to theacid-base method. This method reacted strongly, environmental pollution, easy todamage the product structure.In this paper, based on the characteristics of microbial fermentation and enzymepreparations, try using fermentation and enzymatic to extract tussah pupa skinchitosan. Adjust and optimize the process of traditional chemical method, andcompared of chemical and biological methods.The test result: Use the alkaline-acid method, the purity of chitin and chitosanwill be higher. Use of Lactobacillus fermentation tussah pupa skin can get chitosan,its purity above73%. The fermentation conditions is32℃, the concentration ofglucose10g/100ml, raw volume5g/100ml, inoculum size8%, fermentation period36h. Use of enzymatic treatment tussah pupa skin can get chitosan, its purity of over80%. The enzymatic decalcification condition:80℃,10%citric acid for2hours. Thedeproteinized condition: Alkaline protease treatment materials. Enzyme dosage3000U/g,60℃, pH8,6h.By comparison, summarizes the different methods characteristics of extractedtussah pupa skin chitosan, and the future direction of research and development. Onindustrial production, the traditional acid method is still the first choice. Fermentationand enzymatic methods studies need to be further optimized to form industrialproduction, the creation of environmental protection, ecology, new tussah pupa skinchitosan extraction process.
