Tieguanyin made in Anxi (Fujian, China) was regarded as experiment subject. Thearoma components from tieguanyin in different manufacturing procedures andprocessing techniques and the biological activities of aroma components fromtieguanyin were investigated. This main work was as follows.First, the aroma components of tieguanyin at different releasing time wasextracted by solid phase micro extraction (HS-SPME) and analyzed by gaschromatography-mass spectrometry (GC-MS). The volatiles obtained from theadsorption of XAD-2resin, nano-SiO2, molecular sieve, activated charcoal, silicagels were investigated by GC-MS. It was indicated that the total content of aromacomponents and the content of characteristic flavor compounds were decreasing withthe increased releasing time at50oC and65oC, while the contents of aromacomponents of high boiling point such as farnesene increased at first and thendecreased with the increased releasing time at35oC. Sixty-seven compounds wereidentified from volatiles desorbed from molecular sieve. The varieties of compoundsdesorbed from molecular sieve were the most among five adsorbents.Second, the aroma components from the manufacturing procedures of piling,fixing and baking, the made tea and tea stems of tieguanyin were extracted byheadspace-solid phase micro extraction (SPME) and qualitatively and quantitativelyanalyzed by GC-MS. Twenty-two, thirty-two and nineteen aroma components wererespectively identified in procedures of piling, fixing and baking, and among these,2-ethyl hexanol,2-ethenyl-1,1-dimethyl-3-methylidene cyclohexane, benzyl cyanide,decanal were all detected in these procedures. Meanwhile, sixty-five and fifty-sevencompounds were identified in the made tea as well as tea stems of tieguanyin,respectively, and among these, there were forty identical compounds such asnerolidol, indole, farnesene, benzaldehyde and styrene were found in both sampleswhich accounted for75.70%and86.71%of total content, respectively.Third, the aroma components from tieguanyin of lingering charm type, eliminategreen type and sour fragrant type were analyzed by GC-MS. Fifty-two compounds, sixty compounds and sixty-nine compounds were identified respectively, amongwhich thirty-nine compounds such as nerolidol, farnesene, indole, ocimene, benzylcyanide and hexanoic acid,hexanoate were found in three types of tieguanyinsamples.Finally, the antitumor activity of aroma components from tieguanyin was explored.Aroma components were absorbed by DMSO for the preparation of test sampleswhich were analyzed by GC-MS and antitumor models in vitro were constructedbased on A549. Antitumor activity of sample was detected by the assay of3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT), acridineorange (AO) staining and acridine orange-ethidium bromide (AO-EB) staining.Moreover, cellular morphology and cell apoptosis were observed by LCM and theeffect of samples on cell cycle was investigated by FCM. These results revealed thatthe components from sample were made of naphthalene homologues, benzenehomologues and conpounds such as α-farnesene, ocimene, isolongifolene, and indole.The aroma sample can reduce cell viability and induce the apoptosis of A549.
