Ricin is a kind of virulent toxin from castor seeds, which plays an important role in medicine, agriculture and military affairs fields and has become research focus of anti-cancer drug currently. The traditional extraction way of using buffer, for the long time, partial protein activity reduced. In industrial fields, castor oil was separated in the condition of high voltage. On this condition, the protein deactivitied. For the problems in the process of extraction, in this paper debinding was discussed, and ultrasound wave was used to extract ricin from castor seeds. Extraction rate of ricin was improved, at the same time, the extraction time was shortened. At last, Sapharose-4B was used to purify ricin.Removing grease from castor seeds Leaching was used to remove grease. The degreasing effect of four kinds of organic solvents (petroleum ether, diethyl ether, acetone, ether-alcohol) were compared. And the result showed that acetone was the best solvent.Optimized conditions for ricin extraction process The Box-Behnkens and response surface method were used to optimize and the optimal condition were as followed:extraction time22min, liquid ratio67mL/g, pH7.4, ultrasonic power160W. Under this condition, the extraction yield of ricin reached84.06%.Ricin optimization of purification process In the process of purifying ricin, orthogonal design of experiments L9(3~4) was used to optimize and the optimal condition were as followed:buffer pH7.2, buffer concentration0.15mol/L, D-Galactose concentration0.15mol/L, elution speed1.0mL/min. Under this condition, the extraction yield of ricin reached11.85%and purity was95.10%(HPLC).
