Cadmium (Cd) is a kind of important industrial and environmental pollutants. With the increasing accumulation of Cd in the food chain in vivo concentration, it caused serious threat to the health in both human and animals, especially some of the wild birds and poultry in the environment. Liver was the main target organ, where is the accumulation and exerts toxicity by Cd. Cd also caused damage to hepatocytes, apoptosis and necrosis. In this study, hepatocytes treated by Cd of chicken was taken as a model, and interferenced autophagy process by3-MA and Rap. Then detected the cell activity, antioxidant function, endoplasmic reticulum stress, autophagy and other related indicators. The results showed that:1. Observe from cell viability, morphology and ultrastructure.Show that exposure12h, Cd dose is less than1.0μM, the activity of the liver cells did not change significantly.Doses greater than2.5μM and over24h, the activity of the liver cells in a dose-dependent decrease in dose.These showed there is a anti-injury mechanism from low-dose Cd in hepatocytes, and a toxic effect in high-does.2. The change of antioxidant functions related indicators like T-AOC、GSH-PX、T-SOD、Zn Cu-SOD、CAT、GSH、MDA、ROS、H2O2、·OH in hepatocytes were changes caused by Cd, showed Cd induced hepatocytes damage from oxidative stress.3. Cd can interfere the shear of XBP-1and the mRNA expression of GRP78, ATF4, ATF6, turned out it can induce endoplasmic reticulum stress, play a role in cellular damage.4. The number of autophagosome and autophagic vacuoles were increased by Cd, the mRNA expression of autophagy marker molecule Beclinl、P62、ATG3、ATG5、ATF3and ATG9were interfere by Cd, Indicated Cd induce hepatocyte autophagic.5. Through the establishment of autophagy interference model with autophagy inhibitor3-MA and revulsant RAP, In1.0μM group, detected the autophagic vacuoles, the mRNA expression of autophagy marker molecule Beclinl、P62, and endoplasmic reticulum stress marker molecule GRP78、 ATF4、ATF6and XBP-1.Turned Cd can induce hepatocyte autophagic via Endoplasmic Reticulum stress, uncovered the autophagy play the key role in resist damage caused by low concentrations of Cd in hepatocytes.
