Surface Enhanced Raman Spectroscopy has been considered as a powerful analyticaltechnique in the last years. This thesis mainly focused on the SERS detection of the moleculeswhich are poisonous and have been added illegally in food or fed to the animals, interact weaklywith the SERS substrate. The SERS substrates were modified with particular molecules to trapthe target molecules based on the special intermolecular forces, after which the determined got tothe SERS enhancement area and achieved the SERS enhancement effect. First of all, a Auhemisphere-like structure obtained by evaporating gold on Anodic Aluminum Oxide templateswas served as the SERS substrate; Secondly, for animal remedy Malachite Green, the limit ofdetection could be dropped by optimizing the experiments of the Raman detection; Thirdly,dodecanethiol was employed to function the gold substrate for the SERS detection ofSudanâ… based on the special intermolecular forces-hydrophobic interaction; Finally, the goldSERS substrate was modified with melamine which can form hydrogen bond with clenbuterolhydrochloride, and realized the detection of the target molecules. Against the number ofmolecules whose interaction with the SERS substrate is weak, this method can be used widely invarious fields.This paper is devoted to the preparation of a new type of SERS substrate, which is simplyprepared, highly sensitive and could be used for the qualitative and quantitative SERS detectionof MG, Sudanâ…  and CB in poisonous and harmful materialsbased on the special intermolecularforces. The main content this thesis talking about includes the following four aspects:(1) The preparation of the SERS substrate was exploring first: By the method of vacuumevaporation of Au on the highly ordered porous Anodic Aluminum Oxide templates, thecomplementary structure of which was obtained and used as the SERS substrate.(2) The SERS detection of MG: For the adsorption ability of MG to the substrate is strongbecause of its special Nitrogen structure, there is no need to modify the substrate; After a largenumber of experiments, the optimal experimental conditions were determined, including theexcitation wavelength, the solvent used to prepare the MG, and the pH values of solution, and thequantitative SERS detection of MG was proceeding under those optimal conditions.(3) The SERS detection of Sudanâ… : By changing the excitation wavelength and pH valueof the solution, the Raman intensities of Sudanâ… were observed; The substrate was modified with dodecanethiol for the affinity of Sudan â… o n the gold substrateis weak, and Sudan â…  wouldbe captured with the help of the special intermolecular forces-hydrophobic effect, realizing theSERS enhancement effect.(4) The SERS detection of CB: In the first place, the ability of which to adsorb the substratewas tested; After changing the pH value of the solution, the Raman intensity and peaks positionof CB were observed; For the affinity of CB to the substrate is weak, through the modification ofthe substrate with melamine, and with the help of the special intermolecular forces-hydrogenbond, CB could reach the SERS enhancement area of the substrate.
