The strain of Streptomyces Avermitilis NEAU1069-3used in this experiment was obtained through the treatment of the spores of Streptomyces avermitilis NEAU1069with UV and N-methyl-N’-nitro-N-nitrosoguanidine. From the perspective of drug discovery, mutant strains, in theory, may contain new genes and promising new products which not only provide new chemical scaffolds for drug synthesis, but also force us to understand the biosynthesis mechanism of nature. At the same time, we carried out a systematic research for the metabolites of the strain Streptomces sp. NEAU-Z4, one bioactive compound was isolated and identified so as to provide reference for the drug research and development.Thirty liters broth was given by Large-scale fermentation of the Streptomyces Avermitilis NEAU1069-3.9compounds (1-9) were isolated and purified by using Diaion HP20column, preparative HPLC and semi-preparative HPLC and other chromatographic means. Then, the structure of the compounds were determined on the basis of spectroscopic analysis, including1D and2D NMR, as well as UV, IR, HRESIMS,1H-1H COSY, HMQC, and HMBC. At last, we found that2compounds are novel compounds,25-cyclohexy1-Deo-Phenolic-AVM B1-MS (1) and25-cyclohexyl-Deo-AVM A2-MS (2). The remaining7are known compounds,25-cyclohexy-Avermectin A1(3), Avermectin A2b (4), Avermectin A2a (5),25-cyclohexy-Avermectin A (6), Avermectin A1b (7),25-cyclohexyl-Deo-Phenolic-AVM B1-AGL (8), Avermectin A1a (9)=Streptomces sp.NEAU-Z4was fermented with the volume of30liters, the fermented product after pretreatment was further isolated by Diaion HP20column, ODS column chromatography, silica gel column chromatography, preparative HPLC and semi-preparative HPLC and other chromatographic means. Then, the structure of the compounds were determined on the basis of spectroscopic analysis, including1H-NMR,13C-NMR, IR, UV, HRESIMS,’H-’H COSY, HMQC, and HMBC, and4compounds were finally obtained,2new compounds were named as: Dodeca-8,10-diene-1,3,5-diol (10) and3-(2-hydroxyethyl)indolin-2-one (11), the2known compounds were assigned as N-(4-hydroxyphenethyl)acetamide (12) and N-acetyl-oxotryptamine (13). The compound11express activity against human prostate cancer PC-3with IC5059.5μg/ml.
