Phaffia rhodozyma JMU-MVP14is an astaxanthin over-producing strain gainedthrough mutation breeding by our laboratory, with good prospect for industrialapplication. HPLC combined with open column chromatography were used to studythe characteristic of carotenoids in this strain. In addition, antioxidant activity andeffect on enzymes of the extractions were also studied in vitro. The main resuls wereshown as the following.(1) Antioxidant potential of extracts was influenced significantly by methods ofcell-well disruption and solvents. Compared to extracts of Phaffia rhodozyma treatedwith hydrochloric acid, extracts of Phaffia rhodozyma treated with sodium hydroxideshowed obviously higher antioxidant activity. Methanol was better solvent for theantioxidant component of Phaffia rhodozyma JMU-MVP14.(2) Correlation analysis was done to analyze the relation between totalcarotenoids contents of different extracts and ABTS+scavenging ability, ferricreducing/antioxidant power were significant (the correlative coefficients were0.9765and0.9616, respectively). While correlations between total carotenoids contents andDPPH,·OH scavenging ability were not significant (the correlative coefficients were0.1185and0.5794, respectively). Antioxidant potential of extracts was not onlyrelated to total carotenoids contents, but also the composition of carotenoids.(3) Carotenoids were separated better with MgO and silica gel columnchromatography, with9individual carotenoids separated from the extracts. Six of theindividual carotenoids possessed a carbonyl group in the molecule, conjugated withthe polyene chain skeleton.(4) Extraction-recrystallization was better purification method for astaxanthin inPhaffia rhodozyma JMU-MVP14, with the purity of95%.(5) Astaxanthin purified from Phaffia rhodozyma JMU-MVP14was a kind ofgood scavenging agent for radical, and showed perfect scavenging activities againstDPPH、·OH、ABTS+, with EC50of22.40μg/mL,12.62μg/mL,5.75μg/mLrespectively. Astaxanthin was also an effective inhibitor to acetylcholine esterase andpancrelipase, reached the highest inhibitory of58.28%,74.67%at the concentration of34.78μg/mL、5.00μg/mL, respectively. Astaxanthin also showed slight promotionto tyrosinase, with the highest promotion of29.60%at the concentrate of26.67μg/mL.In this paper, the component together with bioactivities of carotenoids of Phaffiarhodozyma JMU-MVP14were studied, and an effective purification process was alsoestablished. All these results will present a strong support to the utilization ofcarotenoids of Phaffia rhodozyma JMU-MVP14.
