Ruthenium polypyridyl complexes not only have rich optical properties and structureof diversity, but also contain strong ability of binding with DNA. Therefore, thesecomplexes have been applied extensively in field of photochemistry and biochemistry, suchas photoelectric materials, nucleic acid probe, and anticancer drugs. In this thesis, a varietyof new ruthenium complexes were synthesized, and the mechanism of interaction betweencomplexes and DNA were investigated by fluorescence, UV-Vis absorption, viscosity, andgel electrophoresis.With1,10-phenanthroline and benzil derivatives as the raw materials, BFPP, BCPPand BBPP as main ligands have been synthesized. The complexes were synthesized throughabove ligands and cis-Ru(bpy)Cl2·H2O, and the structure of these complexes were analysedby X-ray diffraction. The UV-Vis and fluorescence spectrum of the complexes werecollected under the acetonitrile solvent. And the interaction between the complexes andDNA were study by UV-Vis absorption, fluorescence, the viscosity and gel electrophoresis.The binding capacity between the complexes and DNA increased as follow as[Ru(bpy)2(BFPP)]2+,[Ru(bpy)2(BCPP)]2+,[Ru(bpy)2(BBPP)]2+. The research resultsshowed that introduction of halogen atoms in the the main ligand enhanced the interactionof the complexes with DNA molecules. The gel electrophoresis indicated that the complexescould make cleavage effectively to plasmid PBR322.With2,2’-dipyridine, hydration ruthenium trichloride and benzaldehyde as the rawmaterials, cis-Ru(dnbpy)Cl2·H2O, cis-Ru(dmobpy)Cl2·H2O, cis-Ru(ddbpy) Cl2·H2O andmain ligind2,3-diphenylpyrazino[2,3-f][1,10]phenanthroline(dptatp) have been synthesized.The new complexes were synthesized through above complexes and ligand (dptatp). TheUV-Vis and fluorescence spectrum of the complexes were collected under the acetonitrilesolvent. The interaction between the complexes and DNA were investigaed by UV-Visabsorption, fluorescence, the viscosity and gel electrophoresis. The binding capacitybetween the complexes and DNA decreased as [Ru(bpy)2(dptatp)]2+,[Ru(dbbpy)2(dptatp)]2+,[Ru(dnbpy)2(dptatp)]2+,[Ru(dmobpy)2(dptatp)]2+. The research results showed thatonly the complex [Ru(dmobpy)2(dptatp)]2+presented the significantly fluoresceenceintensity increasement when interacted with DNA. So this complex can be potentiallyapplied as optical switch of nucleic acids. The gel electrophoresis indicated that thecomplexes could make cleavage effectively to plasmid PBR322.
