Morus alba L is the dried leaves of Moraceae plant mulberry, which planted in most ofthe country, was first set out in the "Shen Nong’s Herbal Classic". It can reduce the bloodpressure and lipids, anti-inflammatory and other effects. Evacuating of wind-heat, clearingand moistening the lungs, cleaning liver and improving eyesight are all the main function ofit. In1993, the Ministry of Health determined the mulberry was both food and medicine.In this paper, the Morus alba L was used as study material and the polysaccharideswere studied in the following respects:extraction process, purification technologies andbiological activity. The main experiment results were as follows:1The extraction of polysaccharide of mulberry leaf.Water extraction and alcohol precipitation method for the extraction of mulberry leaves,according to the phenol-sulfuric acid method, measured at490nm absorbance valuepolysaccharide yield. Cellulase, microwave, ultrasound, ultra-fine powder and other methodsof assisted extraction of polysaccharides mulberry leaves, the results show that these fourmethods of extraction rate of mulberry leaves have a more significant effect of improving;simultaneously using orthogonal method of Test for cellulase extraction conditions wereoptimized and the optimum extraction conditions: PH6.0, extraction temperature50℃,enzyme concentration of1.5%, extraction time50min.2The separation and purification of polysaccharide of mulberry leaf.After water extraction and ethanol precipitation to get Mulberry polysaccharide PML1,by measuring the purity of15.44%polysaccharide know. Separation and purification ofPML1using SephadexG-100gel filtration dextran, deionized water as the eluent, to controlthe elution rate of2mL/min, receiving the effluent, and phenol-sulfuric acid solution wasmeasured for each tube receiving method absorbance values plotted elution curves. Peakscollected intermediate portion, polysaccharides combined eluent was concentrated andlyophilized to obtain the purified polysaccharide samples PML2, by measuring the purity of35.94%polysaccharide know..3Biological activity of polysaccharide of mulberry leaf.Of different purity of mulberry leaves were studied in vitro antioxidant activity.Determination of the purity of the two polysaccharides scavenging capacity and reducingpower of its value against DPPH, hydroxyl radical (OH) radicals. Meanwhile, thedetermination of mulberry leaves for α-glucosidase inhibition. The results showed that:Mulberry polysaccharide better clear DPPH, hydroxyl radical (OH) capacity and reducingpower strong, PML2clear and restore capabilities significantly better than PML1.Mulberry polysaccharides on α-glucosidase inhibition, but PML1inhibition than PML2, description ofmulberry leaves may not be able to suppress the α-glucosidase.The results for the development of mulberry leaf polysaccharide, as a kind of oxidationresistance of functional food and medicine has important theoretical and practicalsignificance.
