Cyproheptadine hydrochloride belongs to the tricyclic antihistamine,and it used to treat eczema,urticaria and other allergic disease in the clinical.Studies have also proved that cyproheptadine hydrochloride has the function of promote appetite.So there are some illegal person add cyproheptadine hydrochloride into the animal feed in order to achieve the purpose of increase the animal body weight.Because eating the cyproheptadine hydrochloride remains animalistic food does harm to human health,thus establish a rapid and effective detection method of residual cyproheptadine hydrochloride has shown extraordinary significance on the standardization production of the animal feed and elimination of food safety problems in timely.We intend to establish a new immunological detection method which can testing the concentration of residual cyproheptadine hydrochloride,this article is mainly about the synthesis of cyproheptadine artificial antigen.Cyproheptadine hydrochloride were modified and designed on the basis of preknowledge of artificial antigen synthesis, and several methods were used to insert a series of connecting arm on the cyproheptadine’s different sites. Experiment can be divided into two sections.One is: cyproheptadine directly reacts with anhydride,inserts a connecting arm on the cyproheptadine’s aromatic nucleus sites.The other is:First achieving N-oxide cyproheptadine by oxidating cyproheptadine.Second achieving N-Demethylation of cyproheptadine by the N-oxide cyproheptadine reacts with reduction agent.Last there is a nucleophilic substitution reaction on the piperidine ring in N-Demethylation product of cyproheptadine with drawing a connecting arm which its end with carboxyl group into it.Coupling the semi antigen with carrier protein to get antibody,therefore establish a new immunological detection method which can testing the concentration of residual cyproheptadine hydrochloride.The structures of the target product were characterized by1H-NMR,13C-NMR,MS and IR.
