Combining the protein and polymer to form a new giant molecule called protein/polymerconjugates.When a functional polymer is combined with a protein which have the biologicalactivity, this protein/polymer conjugates would have the dual characteristics of proteins andpolymers. Proteins and polymers both complement each other, which greatly broadens therange of applications of proteins and polymers.pH-Responsive polymer can change its form in different pH environment. Their specialproperties can be widely used in controlled release for drugs, material separation,immunoassay and other fields. In this artcle we synthesized pH-Responsivepolymer-Polyacrylic acid (PAA) via atom transfer radical polymerization (ATRP) by one step.Then PAA is connected with bovine serum albumin (BSA) by covalent bond, and obtainprotein/polymer conjugates. The assembled form is examined in different pH environments.The conjugates can be used as controlled release for drugs.This thesis is divided into three chaptersChapter1: The research of protein/polymer conjugatesThis chapter first reviews the pH-responsive polymers, proteins and protein/polymerconjugates. Then the bonding methods of protein and polymer, advances in proteinself-assembly and protein/polymer conjugate self-assembly are introduced. At last, weintroduce the atom transfer radical polymerization (ATRP) technology and the main work ofthe paper.Chapter2: Preparation of sodium polyacrylate by atom transfer radical polymerizationtechnology(ATRP)Sodium polyacrylate(PAANa) is prepared by one step via atom transfer radicalpolymerization (ATRP) in a mixed solution of aqueous media and N,N-dimethylformamide(DMF). The effects of temperature, pH and VC on the reaction are investigated.The resulting products are characterized by1H-NMR and GPC. The results show that it getsPAANa with the number-average molecular weight6.8KDa and the polydispersity2.4.Chapter3: Synthesis of protein/polymer conjugates and research of its self-assemblybehaviorGT-BSA/PAA is synthesized with BSA and PAA by―grafting to‖technology. Theproduct GT-BSA/PAA is characterized by sodium dodecyl sulfate polyacrylamide gelelectropheresis (SDS-PAGE), native polyacrylamide gel electrophoresis (Native-PAGE),transmission electron microscope (TEM), infrared spectrum (IR) and matrix-assisted laserdesorption/Ionization time of flight (MALDI-TOF). Ultraviolet spectrum (UV), scanningelectron microscope (SEM) and transmission electron microscope (TEM) are used to test its self-assembly behavior. The results show that BSA and PAA are successfully combined bycovalent bond. The combination GT-BSA/PAA exhibits good characteristic of assembly.
