Immunoassays are widely used in many fields ranging from medical diagnostics toenvironmental monitoring. The principal advantage of immunoassays is the specificity ofdetection, which derived from the selection of antibody-antigen binding. Chemiluminescenceenzyme immunoassay (CLIA) has been intensively used in antigen-antibody detection, due to thesensitivity of CLIA is much higher than that of the traditional enzyme linked immunosorbentassay (ELISA). CLIA is a clinical diagnostic method with the basic principle ofradioimmunoassay. It possesses both the advantages of chemiluminescence and immunoassaywith high sensitivity and high selectivity, respectively. Quantum dots (QDs) are semiconductornanoparticles, with high emission quantum yield, narrow emission spectra and size dependentemission wavelength, exhibiting excellent optical properties.Chapter1The principles and applications of immunoassay are reviewed. According to differentlabels, two categories of immunoassay were described. And this reaction mechanism andapplication of those two categories were described in detail. Papers were cited.Chapter2A micro-plate based chemiluminescent immunoassay for measurement of17β-Estradiol in human serum was built. Analytical sensitivity of this method was5.94×10-3μg L-1,the inter-and intra-assay C.V. were below10%, the recovery was between90-100%, and theanalytical result of serum sample was linear to the result of foreign kits.Chapter3A micro-plate based chemiluminescent immunoassay for measurement of cardiactroponin T in human serum was built. Analytical sensitivity of this method was1.6μg L-1, theinter-and intra-assay C.V. were below10%, the recovery was between90-100%, and theanalytical result of serum sample was linear to the result of foreign kits. Chapter4A fluorescence quenching immunoassay was built for analysis myocardial diseasesspecific antigen in serum. The method is easy to operate and with good specific.
