Firstly, a well-defined assembled bead(AMB)with topographies wasprepared with silica colloid as shell beads and hybrid seeded emulsionpolymerized magnetic microspheres as core beads. The immoilized shellbeads coverage of this as-synthesised bead could be adjusted by electrolytein the reaction media. A core-shell magnetic bead(CMB)was prepared asa control bead with a smoonth surface and similar physicochemicalproperties. Then both beads were utilized as biomolecule carriers todetermine the effcet of topological surface on protein interaction. Itshowed no distinguishable difference between AMB and CMB whencoupling small/middle size biomolecules. However, when acted as a carrierof large immune complex, the AMB displayed higher signal intensity,wider detection range, excellent anti-nonspecific protein adsorptionproperties as well as better detection performance. A further research wasmade to determine the improvement of AMB as a carrier in in vitroimmunoassay. The result showed that the AMB with a moderateimmobilized amount of shell beads and streptavidin displayed the mostoptimized detection properties.
