-cyclodextrin (-CD) is cyclic oligosaccharides built up from1,4-glucopyranose unitsthat exhibit a torus-shape structure with a hydrophobic cavity and a hydrophilic exterior.-CD with seven glucose units is known to form inclusion complexes with a variety ofcompounds with suitable size in water through host-guest interaction. Meanwhile,-CD withgood biocompatibility can increase the solubility of lipophilic drugs in water and cover somevery unpleasant smell of the drug itself etc. It can also improve the bioavailability of drugsand reduce their side effects.Both linear and star-shaped poly(glycidyl methacrylate)s (PGMA) were synthesizedthrough atom transfer radical polymerization (ATRP), and functionalized withmono(6-(diethylenetriamine)-6-deoxy)--CD and diethylenetriamine to obtainCD-DETA-PGOHMAs and DETA-PGOHMAs, respectively. The resulting polymers werecharacterized by1H NMR, FT-IR, elemental analysis and TGA, and then used to preparePECs with insulin. The association efficiency and loading capacity of CD-DETA-PGOHMAswere higher than those of DETA-PGOHMAs. The release of insulin depended on theintroduction of-CDs, the backbone architecture of the polymers, as well as the pH condition.The competitive binding release study indicted that insulin could be released rapidly when1-aminoadamantane hydrochloride (ADA) was used as competitive binding guest molecule.The in vitro cytotoxicity study revealed that CD-series polymers have much lower toxicity ofthan D-series. The CD-DETA-PGOHMA/insulin complexes, with lower cytotoxicity andproper release rate, showed great potential for insulin controlled release.Hydrophobic azobenzene conjugated star-shaped PGMA (AZO-PGMA) was synthesizedby click reaction. The structure and photo-responsive property of AZO-PGMA wascharacterized in detail. CD-DETA-PGOHMA and AZO-PGMA can form nanocomplex due tothe interaction between the CD and azobenzene. Their Optical response and reversibilityproperty of nanocomposites was also studied.
