Non-cooked starch is called as raw starch. Traditional technique for starch processing needs gelatinization, liquefaction and saccharification. While raw starch glucoamylase can hydrolyze starch to glucose directly. So it is necessary to set up novel technology of enzymatic saccharification of raw starch without heating, which is important from the viewpoints of energy saving and effective utilization of the biomass. The aim of this paper is to select one strain with raw starch-digesting activity from the soil.Raw starch-digesting strains have been selected using plate hydrolysis spot method and designed as010#, there is large hydrolysis spot in plate with low starch concentration. The optimal culture condition for the cell growth was determined. The optimum concentration of raw starch is5%, the best nitrogen source for growing is NaNO3and the optimum concentration is0.3%.The result showed that the optimum culture temperature is30℃and pH is7.The preliminary study result of raw starch-digesting amylase showed that enzyme can be found both in fermentation and on the surface of raw starch, but it also need to be further studied.
