Two kinds of preparation methods (SMB and DAC) were used to separate4kinds of C-glycosyl flavone (orientin, isoorientin, vitexin, isovitexin) from Bamboo leaf flavone (BLF-d24)and the industrial application prospect of the two technologies were analysised. At the same time, by enzymatic acylation modification of orientin separated and isoorientin, the study focused orientin laurate of radiation protection ability.The main research contents and achievements are summarized as follows:1. SMB and DAC (DAC-100ID100mm) can effectively realize the separation of four kinds of flavone C-glycosides bamboo leaf flavone from BLF-d24, and the sample separation ability is respectively34mg/h and1.25g/h. It can be seen that the production capacity of DAC separation technology is stronger than SMB. DAC preparation system uses ethanol/water as mobile phase, which has the advantages of non-toxic, environmental protection and low cost, compared with SMB preparation system (acetonitrile/water as mobile phase).2. The immobilized lipase (CALB) was used by enzymatic acylation modification of orientin and isoorientin monomer respectively, we obtained orientin laurate and isoorientin laurate. The access of fatty acids makes the vitro antioxidant activity of acylation products decreased. But the evaluation of Rancimat method indicated that esterification products significantly enhance the antioxidant activity in oil system.3. This study used normal human skin fibroblasts (GM0639) as the research object in cell irradiation experiment. We carried out exploratory study of radiation protective effects of orientin laurate by MTT method, detection of free radicals, DNA damage protection.12C6+ion beam (2Gy) irradiation experiments indicated that orientin laurate had a strong protective effect on acute radiation injury effect but lower effect in the process of repair after injury. Free radical detection results showed, orientin laurate had free radical scavenging ability and the protection effect of cell damage caused by the free radical. DNA damage protection tests showed orientin laurate can provide effective protection of GM0639cells in different radiation source (X ray or12C6+ion beam), and the protective effect of12C6+ion beam was better than X-ray (DMSO as positive control).At the same time, orientin laurate had no obvious cytotoxicity to GM0639, so the flavone C-glycosides acylated product can be used as natural radiation protective agent for futhur research and development.
