| In recent years, the development direction about chromatography column is column with high efficiency, high flow rate and high working pressure. The flow conditions in column have been the important factor. Generally, column heterogeneity can decrease column efficiency column axial heterogeneity. By contrast, column heterogeneity has been a hot issue in chromatography field.Generally, the flow condition in column is influenced by the characteristics of column bed. In this work, the effects of column bed characteristics on column axial heterogeneity were studied. First, the effect of column bed multipath characteristics on column axial heterogeneity were studied by computational fluid dynamics method, an UDF program was used to simulate the multipath distribution in column, the results show that multipath distribution can lead to column axial heterogeneity. When multipath distribution degree is larger than0.6, column axial heterogeneity is obvious. The influences of column bed radial heterogeneity on column axial heterogeneity were discussed by number calculation method. Finally, the side wall detect column and glass beads, agarose gel particles were used to studied the effects of particle diameter and particle pore structure on column axial heterogeneity. The results demonstrated that column bed characteristics such as multipath condition, radial heterogeneity, particle diameter and particle pore structure can lead to column axial heterogeneity. Large multipath degree, large radial heterogeneity degree, large particle diameter and large particle surface pore structure can lead to large column axial heterogeneity degree. |
