Preparation And Amplification Of Type ⅡCollagen Hydrolyzate

The efficacity of collagen hydrolyzate function by well understanding and developing, its field of application has also been expanding continually. Normally type Ⅱ collagen exists in animal cartilage collagen, such as chicken cartilage. The hydrolyzate of type II can be used for the treatment of rheumatoid arthritis, but there is little reference about controllable preparation of hydrolyzate. Therefore, this paper taking chicken cartilage as raw material to learn the methods of which controllable type Ⅱ collagen hydrolysate preparation, intending to lay a foundation for its application in medical field.Firstly we have studied the thermal degradation reaction of type Ⅱ collagen, and discovered the regular pattern of thermal degradation, then found a high degree of degradation of suitable preparation conditions.Secondly based on the above studies, this paper gave a further examination about degradation type II collagen under the effect of alkaline protease pyrolysis products of degradation reaction, and revealed the laws of the reaction. Through the comparative research on the degradation products of molecular weight, we obtained the degradation of the alkaline protease suitable conditions. Under the condition of the appropriate enzyme, type II collagen hydrolysate was approached and its relative molecular mass is2450Dalton. The product is clear and transparent (T620nm97.41%), the extraction yield of8.65wt.%.Finally on the basis of laboratory small scale (0.2L), we adopted the method of gradually enlarged, type II collagen hydrolysate preparation of pilot magnification (500L) and the industrialization (3000L) was in progress. In the acquisition of the product of the number-average relative molecular weight smaller than3000daltons utimately, and other indicators were in accord with standard of QB2732-2005.
