Our country has abundant resources of Capsicum, but most people only eat capsicum fruit, lots of Capsicum annuum Leaves are abandoned, the study has found that Capsicum annuum Leaves can inhibit the activity of a-glucosidase. The structure of people’s diet is high-sugar, high-fat and a high calorie, resulting in high incidence of diabetes mellitus in recent years, which threat to human’s health seriously. Capsicum annuum Leaves now become a fashion vegetable; it has non-toxic side effects. If we can develop a natural anti-diabetic drug from Capsicum annuum Leaves, we can not only greatly enhance the added value of Capsicum annuum Leaves, but also bring gospel to the patient. In this study, the contributions of Capsicum annuum Leaves extract to the effects of postprandial blood glucose control were relatively systematically assessed.In this paper, we use water as solvent, firstly, the optimum extraction conditions of Capsicum annuum Leaves were studied through the single factor experiment and response surface experiments. The results show that when the extraction ratio is1:25, temperature is43℃, extraction time is32mins, and the extraction times is3, AGI active ingredient in Capsicum annuum Leaves have the highest rate.Secondly, the inhibition of Capsicum annuum Leaves and acarbose to different sources of a-glucosidase were compared, the results showed Capsicum annuum Leaves has strong inhibitory activity on a-glucosidas that comes from porcine pancreatic intestinal, yeast and rice, while showes no inhibitory activity on a-glucosidase that comes from bacteria. On this basis, the inhibitory kinetic of Capsicum annuum Leaves to different sources of a-glucosidase were measured. The results showed that the inhibition type of Capsicum annuum Leaves on a-glucosidas that comes from porcine pancreatic intestinal, yeast and rice is competitive inhibition.Finally, we established Caco-2cell monolayer model to study the hypoglycemic mechanism of Capsicum annuum Leaves. First, whether Capsicum annuum Leaves can inhibit the activity of enzymes that are related to digestion of carbohydrates were studied; then whether Capsicum annuum Leaves can inhibit glucose transport under different blood sugar condition were studied. The results show that Capsicum annuum Leaves can inhibit both maltase and sucrase, and also has influence on glucose transporting.
