Baoding is a water-shortage like other cities in Hebei province, which is Located in mid part of province. Water source depend on groundwater in this city. The development of industry and agriculture cause water shortage and contaminated surface water and groundwater. In order to know the pollution status of this city, investigation of difference corporation was done. This paper concentrated on microbial diversity of groundwater beneath a petroleum-refine plant. General situation of microbe and change of microbial diversity in petroleum-contaminated groundwater is scarcity in China. Main research focus on certain species degrading correspond contamination in China .Applying tradition culture-identify technique investigating density of bacteria living in contaminated groundwater, applying molecule microbe technique study spatial-temporal variety of microbial diversity. Constructing clone library of severely contaminated groundwater for sketch view of species type and abundance. DGGE technique investigating microbial diversity change in vertical section and time serial. Microbe community in this type groundwater is uniqueness, the main species is Proteobacteria, this community including 13 species bacteria. Statistics analysis is used to analyze relationship of environmental parameter and microbial diversity, obtaining a optimum temperature scale which microbial diversity is high. The similarity of Difference depth groundwater samples in one well is high, the similarity level of this samples change as time. The metabolism gradually become aerobic from anaerobic, at the same time the normal species appears, this because of bad impermeability of monitor well.
