This paper mainly study a non-confocal Laser Induced Fluorescence (LIF) detection system, which is used to count micro-ball flowed into the channel of the Microfluidic chip.One kind of micro-ball (diameter of 5.6μm) is dyed using red fluorescent dye,the other is dyed using green fluorescent dye. Those of micro-ball with different fluorescent dye can be used in biochemical analyses and biology molecules will react on the micro-ball. When the biology reaction is finished we take LIF detection on the micro-ball and count on the micro-ball attached with two kind of fluorescent dye. By counting the number of the micro-ball we can complete the biochemical analyses. This method can help us to finish biochemical analyses with less reagent, but more simple operation and high efficiency.We study LIF detection of the microfluidic biochemical analyses system. The works we have done as below: the design of whole structure of the non-confocal LIF detection system; the design of the device to couple excitated light into the SMF, transport 532nm and 635nm laser direct to the detection area; designing the device to divide, filter and detect the fluorescence signal. Discuss the method to improve the efficiency of collecting signal and signal-noise ration. Make the setup of detection device, high voltage supply of PMT and low-noise preamplifier; Doing LIF detection experiment to green fluorescent dye. We got the optimization of detection device based on the experiment's result; Giving improving advice for future research.
