Using Tc-inducible CaM/CaMBP transgenic tobacco systems as experimental material, which are newly constructed by our lab, we studied the relationship among CaM, AOS generation and the antioxidant enzyme in vivo.The present research results included three parts:For the first part, we measured the CaM activity of 25 inducible nucleus CaMBP expression transgenic tobacco lines by the improved quantitative assay of plant calmodulin by phosphordiesterase (PDE). We selected four transgenic tobacco lines (8, 9, 13 and 15a) who the activity of CaM was good decreased in seedlings after treatment with Tc, and we found the optimumTc-inducible time.For the second part, we used the four transgenic tobacco lines's leaf diskselected in the first part , a Tc-inducible cytoplasm CaMBP line and a Tc-inducible cytoplasm CaM line as experimental material, measured the change of CaM activity of transgenic tobacco leaf disk in 24 hours after treatment with Tc, and studied the effect of controlling CaM activity on AOS generation. We found that the content of AOS of control HO2020 was almost not changed with or without Tc induction. The H2O2 content are all increased in which CaM expression was raised or reduced; but the O2 ~ content was decreased when CaM activity is raised, and the 02 " content was increased when CaM activity was decreased. These indicate that CaM may be involved in the control of the generation of AOS.In addition, we studied the five antioxidant activity of guaiacol peroxidase (GPX), catalase (CAT), superoxide dismutase (SOD) , ascorbate-4-peroxidase (APX) and glutathione reductase (GR) of the four transgenic tobacco lines which have been used in the second part. We found the change of the five antioxidant enzymes activity were all in accordance with the change of CaM activity, the antioxidant enzyme activity was enhanced when the CaM activity was increased; the antioxidant enzyme activity was decreased when the CaM activity was reduced, while the activities of five antioxidant enzymes of control HO2020 almost had no change with or without Tc induction.All these indicate that CaM can be involved in the control of AOS generation through the regulation of antioxidant enzymes .
