In recent years, IVF and embryo development increasingly goes into the deep, the important basal of these study is mammalian early embryos culture in vitro Consequently,establish mammalian early embryos culture system has been important study question. However, early embryo development block often happened cultured in vitro, the mechanism of developmental block of mammalian early embryos is unclear, surroundings difference of embryo development in vitro and in vivo is oxygen concentration, oxygen concentration in vitro is higher, the metabolism of molecular oxygen in embryos formed ROS, recently, there are a lot of reports on the effect of ROS on early embryo development.By the way of detecting HA concentration of mouse embryo medium, we discuses the effect of ROS on the development block of the mouse early embryos and offered the reliable theories for enhancing the developing efficiency of the early embryos of the mammalian.The 1-cells embryo was gained from the oviduct of mouse superovulated, and was cultured in vitro. We apply ultraviolet-visible spectrophotometry to detecting H2O2 concentration of mouse embryo medium to explore the relationship between embryo development in vitro and H2O2 content.0~24h,24~48h,48~72h,72~96 h H2O2 concentration in embryo medium is 3. 379 μM, 2. 071μM,1.218μM,0.891 μ M respectively.. 0~24 h H2O2 concentration is significantly higher than the other groups, and there were not significant difference between the other three groups;. When embryos are cultured from 0 to 36 hours, the concentration of H2O2 in medium higher, blastula developing rate is lower, shows negative correlation. And correlation coefficient is -0.784.In conlusion: H2O2 concentration is highest in medium before late 2-cell; 036 h, H2O2 concentration in medium have important effect on mouse blastocyst development rate.
