The tomato blue light receptor Phototropins are the protein that have the very function to mediate the hypocotyl's phototropism, elongation, chloroplast movement, leaf expansion, stoma opening and closing and so on. Phototropins are composed of an amino-terminal photosensory domain called LOV (Light, Oxygen, Voltage) that can tightly bound to the FMN(flavin mononucleotide) molecules and a carboxy-terminal Ser/Thr protein kinase domain have the so called light-dependent phosphotylation. Since these genes had very important functions in plants and they had not been reseached enough in tomato, so this experiment designes two pairs of primer for phot-1 and phot-2 RNAi vectors. Because the tomato blue light receptor's genes completed sequence have not been acquired and I need them to construct their overexpression vectors, I have used Rapid amplification of 3' and 5' cDNA ends(RACE) technology to acquire their completed sequence. Nowadays transgenetic tomato usually utilize Agrobacterium tumefaciens to mediae. The recombinate vector transferred to tomato cotyledon via Agrobacterium tumefaciens, then screening the explant with medium having corresponding antibiotic. This method is easily controlled, and it do not need strict experiment condition and the explant's regeneration rate is high, so it has become the most popular method. This experiement also utilize this method, furthermore, the most suitable condition of seeding and transfection have been analysed. |
